Objective

Validate TIGIT–CD155 matched-pair binding (KD, k_on, k_off) and confirm antibody blockade via competition — the two experiments a checkpoint screening campaign runs before trusting hits.

Materials

Biotinylated TIGIT (ECD) — capture format；CD155 ECD — analyte
Streptavidin biosensors（BLI）或 streptavidin chip（SPR）
Running buffer: PBS-T pH 7.4 + 0.1% BSA（防非特异）
Analyte: CD155 ECD（SEC 确认单体/二聚状态）
Blocking reagents: anti-TIGIT / anti-CD155 antibody；DNAM-1（CD226）对照

Procedure

捕获：biotinylated TIGIT 低密度捕获（Rmax 20–50 RU / 0.2–0.5 nm）。
平衡：PBS-T+BSA 至基线稳定。
浓度系列：CD155 2 倍稀释系列（覆盖低 nM 级 KD），≥5 浓度 + 0；结合 180 s，解离 300 s。
再生：10 mM glycine pH 2.5–3.0 短脉冲（10–15 s）；验证 ≥5 循环活性。
竞争实验：CD155 与 anti-TIGIT（或 anti-CD155）预孵育 30 min → 注入 → 结合信号应显著下降；
交叉对照：CD155 与 DNAM-1 的结合对照（区分 TIGIT 特异性阻断 vs 配体阻断）。

Interpretation

文献报道 TIGIT–CD155 KD 为低 nM 级；与 datasheet 差 10× 以上先查密度/缓冲液/二聚。
竞争抑制率 = (1 − 结合信号/对照) × 100%；记录抑制率与对照抗体对比。
DNAM-1 对照：若抗体不阻断 CD155–DNAM-1，说明仅阻断 TIGIT 通路（生物学不同，需报告）。
