3. Why IL-15 (and not IL-2) for NK
- No Treg expansion — IL-2 expands Tregs, which suppress the very effectors you're growing.
- Better NK survival — IL-15 promotes NK homeostasis; IL-2-driven NK cultures show more apoptosis without feeder support.
- Lower systemic toxicity in vivo — relevant if you're scaling toward clinical use.
4. Common failures (and fixes)
- Starvation by under-feeding — IL-15 is consumed and decays; feed every 2–3 days, don't just top up media.
- Serum interference — some sera contain factors that mask IL-15 activity; for potency comparisons use defined media.
- Wrong readout timing — measuring fold-expansion at day 7 vs day 14 tells different stories; fix your endpoint before comparing cytokines.
