Why compare both allotypes?

The V158/F158 polymorphism of CD16a (FcγRIIIa) shifts IgG1 affinity ~2–5× (reported in literature). If you're evaluating an Fc-engineered antibody or an ADCC-enhancing format, the improvement must hold on both allotypes — a candidate that only gains affinity on 158V may fail in the 158F-majority patient population.

Interpretation

  • ΔKD ratio 158V/158F: report as fold-difference (e.g., 158V binds 3× tighter). Compare your value with the supplier's datasheet to validate the lot.
  • Fc engineering readout: plot candidate vs WT-IgG1 on both allotypes; look for parallel shift (affinity gain) vs selective gain.
  • ADCC correlation: binding data ≠ killing; if functional claim needed, add cell-based ADCC (NK or PBMC) with matched donor allotype if possible.

Common pitfalls

  • Avidity contamination: Fc-fusion CD16a or dense surface inflates both allotypes differently → low-density monomeric ECD only.
  • Lot mixing: 158V and 158F must come from the same batch philosophy (same supplier/expression system) — mixing vendors corrupts the ratio.
  • Glycosylation drift: HEK293 vs CHO changes binding; keep expression system constant and stated.
  • Single-concentration "affinity" claims: always full concentration series + global fit.

What to request from your supplier

  • 158V and 158F from the same expression system, allotype-verified (sequencing/MS)
  • SPR KD vs a named reference IgG1 for each allotype
  • Biotinylated versions for identical capture format
